Evolutional and Functional Analysis of T-box Regulon in Bacteria: Identification of New Genes Involved in Amino Acid Metabolism

ثبت نشده
چکیده

Motivation: T-box antitermiantion is the most distributed mechanism of regulation of various amino acids in Gram-positive bacteria. Identification of the T-box regulon and a metabolic analysis of amino acid biosynthesis and transport is one of problems of comparative genetics, genomics and molecular biology. Results: Search for T-box elements and analysis of operon structures identified a large number of new candidate T-box regulated genes, mostly transporters, in Gram-positive bacteria. We assign amino acid specificity for a large number of candidate transporters as well as for other new amino acid related genes. Availability: The program is available by request to the author.

Comparative genomic analysis of T-box regulatory systems in bacteria.

T-box antitermination is one of the main mechanisms of regulation of genes involved in amino acid metabolism in Gram-positive bacteria. T-box regulatory sites consist of conserved sequence and RNA secondary structure elements. Using a set of known T-box sites, we constructed the common pattern and used it to scan available bacterial genomes. New T-boxes were found in various Gram-positive bacte...

The global regulator CodY in Streptococcus thermophilus controls the metabolic network for escalating growth in the milk environment.

CodY is a transcriptional regulator conserved in the low-GC group of Gram-positive bacteria. In this work, we demonstrated the presence in Streptococcus thermophilus ST2017 of a functional member of the CodY family of global regulatory proteins, S. thermophilus CodY (CodYSt). The CodYSt regulon was identified by transcriptome analysis; it consisted predominantly of genes involved in amino acid ...

Overall control of nitrogen metabolism in Lactococcus lactis by CodY, and possible models for CodY regulation in Firmicutes.

CodY, a pleiotropic transcriptional regulator conserved in low G+C species of Gram-positive bacteria, was previously described to be the central regulator of proteolysis in Lactococcus lactis. In this study, over 100 potential CodY targets were identified by DNA-microarray analysis. Complementary transcriptional analysis experiments were carried out to validate the newly defined CodY regulon. M...

Regulation of biosynthesis and transport of aromatic amino acids in low-GC Gram-positive bacteria.

Computational comparative techniques were applied to analysis of the aromatic amino acid regulon in Gram-positive bacteria. A new candidate transcription regulation signal of 3-deoxy-D-arabino-heptulosonate-7-phosphate synthase and shikimate kinase genes was identified in Streptococcus and Lactococcus species. New T-boxes were found upstream of aromatic amino acid biosynthesis and transport gen...

Identification, Cloning and Structural Analysis of Major Genes from Portulaca oleracea L. Hairy Roots that Involved in the Biosynthesis of Dopamine

Dopamine is one of the important medications of Portulaca oleracea L. To optimize the production of dopamine, one of the methods is the identification and engineering of metabolite pathways. To investigate the tyrosine decarboxylase (TDC) and tyrosinase, which seem to be the most important genes in dopamine synthesis pathway, hairy roots were produced from Portulaca oleracea using Agrobacterium...
